چکیده
SOURCE/DESCRIPTION; This cosmid HCOD13 has been isolated by an oligonucleotide (1) from a human genomic cosmid library. POLYMORPHISM: PvuII identifies >10 allelic VNTR polymorphisms with bands between 4.0 and 10.0 kb. TaqI, Rsal Mspl and Pstl identify the same VNTR polymorphism.

DHPLC Applications: Finding DNA Variation on the Y Chromosome
Denaturing High-Performance Liquid Chromatography (DHPLC) is a recently developed technique forthe detection of single nucleotide polymorphisms (SNPs) and mutations. It involves the comparisonbetween two or more DNAs as a mixture of denatured and reannealed PCR products. The methodologyis based on the principle of reversed phase liquid chromatography and uses a unique DNA sepa...
متن کاملIsolation of polymorphic DNA fragments from human chromosome 4.
We have identified and characterized 40 DNA probes detecting restriction fragment length polymorphism (RFLP) on human chromosome 4. Single copy human clones were isolated from a bacteriophage library enriched for chromosome 4 sequences. Each clone was hybridized to somatic cell hybrid DNAs for verification of its species and chromosomal origin and for regional localization. Sequences specific f...
متن کاملUSE OF VECTORETTE AND SUBVECTORETTE PCR FOR THE ISOLATION OF TERMINAL SEQUENCES FROM Y EAST ARTIFICIAL CHROMOSOME (YAC) CLONES
Development of yeast artificial chromosome (Y AC) vectors, molecular cloning of large segments of chromosomal DNA, and their propagation in yeast cells has become feasible. Overlapping Y AC provides a route to the development of physical maps of entire mammalian chromosomes. A rapid method was developed to isolate and sequence termini of Y AC inserts quickly. The Y AC clone is digested wit...
متن کاملMicrosatellite (SSR) amplification by PCR usually led to polymorphic bands: Evidence which shows replication slippage occurs in extend or nascent DNA strands
Microsatellites or simple sequence repeats (SSRs) are very effective molecular markers in population genetics, genome mapping, taxonomic study and other large-scale studies. Variation in number of tandem repeats within microsatellite refers to simple sequence length polymorphism (SSLP); but there are a few studies that are showed SSRs replication slippage may be occurred during in vitro amplifi...
